چکیده

Schneider (1999) recently addressed the question of whether Ca 2 1 sparks arise from the opening of a single ryanodine receptor (RyR) channel or the simultaneous opening of several channels. The discussion highlighted the importance of single RyR channel permeation and gating in the interpretation of Ca 2 1 spark data. The Schneider (1999) perspective inspired us to extend this theoretical discussion by using a published kinetic model of modal RyR gating to actually simulate RyR channel gating and permeation that may underlie a Ca 2 1 spark in cardiac muscle.

Structural determinants of skeletal muscle ryanodine receptor gating.

Ryanodine receptor type 1 (RyR1) releases Ca(2+) from intracellular stores upon nerve impulse to trigger skeletal muscle contraction. Effector binding at the cytoplasmic domain tightly controls gating of the pore domain of RyR1 to release Ca(2+). However, the molecular mechanism that links effector binding to channel gating is unknown due to lack of structural data. Here, we used a combination ...

Topic Introduction Patch-Clamp Electrophysiology of Intracellular Ca Channels

The modulation of cytoplasmic free Ca concentration ([Ca]i) is a universal intracellular signaling pathway that regulates numerous cellular physiological processes. Ubiquitous intracellular Carelease channels localized to the endoplasmic/sarcoplasmic reticulum—inositol 1,4,5-trisphosphate receptor (InsP3R) and ryanodine receptor (RyR) channels—play a central role in [Ca ]i signaling in all anim...

Coupled gating between individual skeletal muscle Ca2+ release channels (ryanodine receptors)

Excitation-contraction coupling in skeletal muscle requires the release of intracellular calcium ions (Ca2+) through ryanodine receptor (RyR1) channels in the sarcoplasmic reticulum. Half of the RyR1 channels are activated by voltage-dependent Ca2+ channels in the plasma membrane. In planar lipid bilayers, RyR1 channels exhibited simultaneous openings and closings, termed "coupled gating." Addi...

Channel Gating Dependence on Pore Lining Helix Glycine Residues in Skeletal Muscle Ryanodine Receptor.

Type 1 ryanodine receptors (RyR1s) release Ca(2+) from the sarcoplasmic reticulum to initiate skeletal muscle contraction. The role of RyR1-G4934 and -G4941 in the pore-lining helix in channel gating and ion permeation was probed by replacing them with amino acid residues of increasing side chain volume. RyR1-G4934A, -G4941A, and -G4941V mutant channels exhibited a caffeine-induced Ca(2+) relea...

Ryanodine receptor gating controls generation of diastolic calcium waves in cardiac myocytes

The role of cardiac ryanodine receptor (RyR) gating in the initiation and propagation of calcium waves was investigated using a mathematical model comprising a stochastic description of RyR gating and a deterministic description of calcium diffusion and sequestration. We used a one-dimensional array of equidistantly spaced RyR clusters, representing the confocal scanning line, to simulate the f...
